Responses of Isolated Esophageal Muscle Strip of Frog and Tortoise to Some Autonomic Drugs, 5-Hydroxytryptamine, and Histamine

Abstract
1. Responses to some autonomic drugs, 5-hydroxytryptamine (5-HT), and histamine were investigated in isolated longitudinal and circular muscle strip of esophagus of frog and tortoise.
2. Longitudinal muscle strip of frog showed rhythmical spontaneous movement circular muscle strip of frog showed irregular movement a part of longitudinal muscle strips of tortoise esophagus showed weak irregular movement.
3. Acetylcholine contracted all the muscle preparations. Atropine blocked the contraction.
4. Sympathomimetic amines (norepinephrine, epinephrine and isoproterenol) inhibited the movement of the longitudinal muscle of frog. They produced inhibition or contraction in the other three preparations. The inhibitory responses were not observed in the presence of pronethalol. The augmentative responses were not observed in the presence of phenoxybenzamine.
5. Nicotine produced contraction followed by inhibition. Hexamethonium blocked the both effects, atropine did the contractile effect.
6. DMPP relaxed the both muscle strips of frog esophagus. The relaxations were not affected by hexamethonium. DMPP produced contraction in longitudinal muscle strip of tortoise and relaxation in circular strip. The responses of the tortoise esophagus to DMPP were blocked by hexamet honium and bretylium. The contractile response of longitudinal¢¥ muscle was prevented by ph enoxybenza mine but not by atropine.
7. McN-A-343 produced contraction of longitudinal muscle strip of frog and the both muscle strips of tortoise. The contraction was inhibited by atropine. McN-A-3-13 ,produced relaxation of circular muscle strip of frog esophagus.
8.5-HT produced contraction of longitudinal muscle strip of fro- and tortoise, produced relaxation of circular strip of them. The stimulant and, inhibitory effects of 5-HT on the tortoise esophageal stripp did not appear¢¥ in the presence of-methysergide.
9. Histamine contracted the circular muscle strip of frog and both musclestrips of tortoise. Promethazine blocked the contractions.
